Effect of LFA-1 and ICAM-1 antibody treatment on murine corneal allograft survival.
PURPOSE To examine the effect of anti-LFA-1 and anti-ICAM-1 antibody treatment on orthotopic corneal graft survival in a mouse model. METHODS Anti-LFA-1 and anti-ICAM-1 antibodies were administered intraperitoneally before and shortly after orthotopic corneal transplantation. Grafts were observed by biomicroscopy, and survival times were determined. Cytotoxic T lymphocyte (CTL) and delayed-ty...
متن کاملPromotion of murine orthotopic corneal allograft survival by systemic administration of anti-CD4 monoclonal antibody.
A mouse model of orthotopic corneal allograft rejection was used to examine the efficacy of anti-CD4 and anti-CD8 monoclonal antibodies in preventing immunologic rejection of corneal allografts. Although it is believed by many that corneal graft rejection is mediated, at least in part, by CD8-positive cytotoxic T-lymphocytes, systemic administration of anti-CD8 antibody did not reduce the rejec...
متن کاملDeletion of the chemokine receptor CCR1 prolongs corneal allograft survival.
PURPOSE Many corneal grafts undergo immune rejection, and current therapies are associated with many side effects. The purpose of this study was to identify critical chemokine pathways involved in generating the alloimmune response to corneal transplants. METHODS Orthotopic corneal transplantation was performed in fully mismatched strains. Cytokine and chemokine receptor gene expression was d...
متن کاملAdministration of an anti-interleukin 2 receptor monoclonal antibody prolongs cardiac allograft survival in mice
Administration of the monoclonal antibody M7/20, which binds to the murine interleukin-2 (IL) receptor, significantly prolongs cardiac allograft survival in two H-2-incompatible strain combinations of inbred mice. The results support the important role of the IL-2 receptor in the mechanism of graft rejection, and suggest its suitability as a target for immunosuppressive therapy.
متن کاملLFA-1-specific therapy prolongs allograft survival in rhesus macaques.
Outcomes in transplantation have been limited by suboptimal long-term graft survival and toxicities associated with current immunosuppressive approaches. T cell costimulation blockade has shown promise as an alternative strategy to avoid the side effects of conventional immunosuppressive therapies, but targeting CD28-mediated costimulation alone has proven insufficient to prevent graft rejectio...
